NAD+ is one of the most important molecules in our bodies, however, levels have been shown to decline with age. Understanding NAD+ metabolism (how it is generated and consumed) can be quite complicated and it’s still being investigated, yet this understanding will help identify ways to rejuvenate levels of NAD+. One of the best ways to navigate the metabolism is to appreciate the structure and nomenclature of NAD+ and its precursors, nicotinamide riboside (NR) and nicotinamide mononucleotide (NMN). As you will learn in this video, by understanding the different pathways through which NAD+ is made and used up, there are three strategies through which NAD+ levels could be rejuvenated.
You can think of the strategies as make, break or take. How you can enhance production (by activating NAMPT), how you can inhibit breaking down (eg. CD38 inhibitors such as apigenin) and how you can take (NAD+ precursors such as NMN, NR, niacin (NR) and NAM). The majority of work has been conducted in preclinical models, however as Eleanor explains, clinical trials are also in progress.
